Enterokinase is a specific protease that cleaves after lysine at its cleavage site Asp-Asp-Asp-Asp-Lys. It will sometimes cleave at other basic residues, depending on the conformation of the protein substrate.

This preparation is purified from Pichia pastoris containing a clone of the light chain of the bovine enterokinase gene.

    1 unit is defined as the amount of enzyme required to cleave 25 µg of a MBP-EK-paramyosin-ΔSal substrate to 95% completion in 16 hours at 25°C in a total reaction volume of 25 µl.

    Two fold dilutions of Enterokinase, light chain are incubated with 25 µg MBP-EK-paramyosin-ΔSal substrate in 20 mM Tris-HCl, 50 mM NaCl, 2 mM CaCl2 (pH 8.0 @ 25°C) in a 25 µl reaction. The reaction mix is incubated at 25°C for 16 hours. Separation of reaction products are visualized by SDS-PAGE.
