产品信息
神经氨酸苷酶是乙酰神经氨酸水解酶(唾液酸酶)的俗称。该酶可催化水解糖蛋白和寡糖的 α2-3、α2-6 和 α2-8 连接的 N-乙酰神经氨酸残基。

该酶克隆自产气荚膜梭菌(Clostridium perfringens),在 E. coli 中过表达。

特性和用法
单位定义
1 单位指 10 µl 反应体系中,37℃ 条件下,5 分钟内将 1 nmol Neu5Acα2-3Galβ1-3GlcNAcβ1-3Galβ1-4Glc-7-amino-4-methyl-coumarin(AMC)中超过 95% 的末端 α-Neu5Ac 水解所需要的酶量。

单位活性检测条件
在 10 µl 1X GlycoBuffer 1 反应体系中,将两倍稀释的 α2-3,6,8 神经氨酸苷酶与 1 nmol AMC 标记底物进行温育。反应混合液在 37℃ 条件下温育 5 分钟。通过薄层色谱法观察反应产物的分离情况(3)。
