  单位定义

  1 个单位是指在 37℃ 条件下，10 µl反应体系中，1 小时内能保护 1 µg λ DNA 不被 AluI 限制性内切酶切割所需酶量。

  反应条件

  1X AluI 甲基转移酶反应缓冲液
  Supplement with 80 µM S-腺苷甲硫氨酸（SAM）
  Incubate at 37°C

  对照保护实验

  AluI 甲基转移酶与 1 µg λ DNA 在 10 µl补充了 80 µM S-腺苷甲硫氨酸的 1X AluI 甲基转移酶缓冲液中于 37℃ 温育 1 小时，后于 65℃ 温育 15 分钟。AluI 甲基转移酶的保护能力测定通过添加 40 µl 补充了 10 mM MgCl₂ 和 10 个单位 AluI 限制性内切酶的 NEBuffer 1，37℃ 条件下温育 30 分钟后用琼脂糖凝胶电泳分析。
