产品信息

虽与 HpaII 甲基转移酶识别序列相同，但 MspI 甲基转移酶修饰 CCGG 序列的外侧胞嘧啶残基（C5）。

大肠杆菌菌株，携带克隆自莫拉氏菌（Moraxella）属（ATCC 49670）的 MspI 修饰基因。

  • 特性和用法

    单位定义

    1 个单位是指在 37℃ 条件下，10 µl反应体系中，1 小时内能保护 1 µg λ DNA 不被 MspI 限制性内切酶切割所需酶量。

    反应条件

    1X MspI 甲基化酶反应缓冲液
    Supplement with 80 µM S-腺苷甲硫氨酸（SAM）
    Incubate at 37°C

    1X MspI 甲基化酶反应缓冲液
    50 mM Tris-HCl
    100 mM NaCl
    5 mM β-ME
    10 mM EDTA
    (pH 7.5 @ 25°C)

    对照保护实验

    MspI 甲基转移酶与 1 µg λ DNA 在 10 µl 补充了 80 µM S-腺苷甲硫氨酸的 1X MspI 甲基转移酶反应缓冲液中，于 37℃ 温育 1 小时，后于 65℃ 温育 15 分钟。MspI 甲基转移酶保护能力的测定是通过添加 40 µl 补充了 10 mM MgCl2 的 NEBuffer 2 和 5 单位 MspI 限制性内切酶。37℃ 条件下温育 MspI 后用琼脂糖凝胶电泳分析。
