NEBNext® 接头采用了一种新型的发卡环状结构,能以更高的效率连接末端经修复和加 dA 尾的 DNA 片段。环状结构 DNA 中包含一个 U,当 U 被 USER® 酶(UDG 和 Endo VIII 的混合物)切掉后,环状结构打开,使它可以成为 PCR 反应的底物。通过 NEBNext Index 引物在 PCR 步骤中引入 Barcode,从而有助于实现多重测序文库制备。此试剂盒包含 96 对预混合的独特的 i5 和 i7 Index 引物,封装在可穿透铝箔密封的一次性 96 孔板中。
NEBNext 接头引物可与 NEBNext 产品以及其它兼容 Illumina 平台的标准文库制备流程结合使用。
还提供双端 Barcode 引物(NEB #E7600、NEB #E7780),以及 12 种和 96 种单端 Index 引物(NEB #E7335、NEB #E7500、NEB #E7710、NEB #E7730、NEB #E6609)。也提供甲基化的 NEBNext 接头(NEB #E7535),可用于重亚硫酸盐测序流程。
流程:
NEBNext 接头专为 DNA、ChIP DNA 和 RNA(但不包括 Small RNA)文库制备而设计,能够实现高效的接头连接和高文库产量,并最大限度减少接头二聚体的形成。NEBNext 接头采用了一种新型的发卡环状结构,能以更高的效率连接末端经修复和加 dA 尾的 DNA 片段。环状结构 DNA 中包含一个 U,当 U 被 USER 酶(UDG 和核酸内切酶 Endo VIII 的混合物)切掉后,环状结构打开,使它可以成为 PCR 反应的底物。通过 NEBNext Index 引物在 PCR 步骤中引入 Barcode,从而有助于实现多重测序文库制备。此试剂盒包含 96 种预混合的 8 bp Index 引物,封装在可穿透铝箔密封的一次性 96 孔板中。
