  1. Storage and stability

 

When stored at 2-30 C , the empty Cell-Free DNA Blood Collection tubes is stable through expiration date.

Shelf life of the empty Cell-Free DNA Blood Collection tubes is two years.

When the blood is collected in the tubes, the Cell-Free DNA in the Boykyo Cell-free DNA Blood Collection tubes can be stable for up to 14 days, and circulating tumor cells are stable for up to 7 days, at room temperature between 6-37 C .

 

  1. Temperature

 

The Boykyo Cell-free DNA Blood Collection tubes can be stored at 2-30 C .

The Boykyo Cell-free DNA Blood Collection tubes can be used at room temperature between 6-37 C .

 

  1. Protocol

 

  • Double spin protocols are commonly used to separate blood plasma for Cell‑Free DNA isolation.

300 x g for 20 min followed by 5000 x g for 10 min or 1600 x g for 10 min followed by 16000 x g for10 min

 

  • The single spin protocol can also be used to separate blood plasma for Cell‑Free DNA isolation.

1600 x g for 15 min

 

3.3  Perform DNA extraction from plasma in accordance with assay instructions or standard laboratory protocols. For example, Plasma cfDNA can be purified using the commercially available QIAamp®Circulating Nucleic Acid Kit (QIAGEN®, Santa Clarita, CA). For optimal results, the manufacturer’s recommended protocol was modified slightly by increasing the duration of the proteinase K treatment from 30 min to 1 hour at 60 C . cfDNA samples were stored at ‑80 C  until analysis by PCR.

  1. Instructions for use

 

5.1  Blood Collection

General guidelines for specimen collection can be found in Clinical and Laboratory Standards Institute (CLSI) H3-A6, Procedures for the Collection of Diagnostic Blood Specimens by Venipuncture1.

5.1.1  Collect specimen (avoid backflow):

  1. Place the patient’s arm in a downward position.
  2. Hold the tube with the stopper uppermost.
  3. Release tourniquet once blood starts to flow in the tube.
  4. Fill tube in 10 ml
  5. Ensure the tube contents do not touch the stopper or the end of the needle during collection.

5.1.2  Remove tube from adapter.

5.1.3  To avoid hemolysis, immediately mix by gentle inversion 8 to 10 times to ensure adequate mixing of the chemical additives with the blood specimen.

  • Plasma Separation

 

5.2.1   Plasma separation centrifugation protocols included the single spin protocol (1600 x g for 15 min) and two double spin processes(300 x g for 20 min followed by 5000 x g for 10 min or 1600 x g for 10 min followed by 16000 x g for10 min)

 

  • cfDNA Isolation From Plasma
